A likely role for a novel PH-domain containing protein, PEPP2, in connecting membrane and cytoskeleton Biocell
Zou,Yi; Zhong,Wenping.
PH domains (pleckstrin homology) are well known to bind membrane phosphoinositides with different specificities and direct PH domain-containing proteins to discrete subcellular apartments with assistances of alternative binding partners. PH domain-containing proteins are found to be involved in a wide range of cellular events, including signalling, cytoskeleton rearrangement and vesicular trafficking. Here we showed that a novel PH domain-containing protein, PEPP2, displayed moderate phosphoinositide binding specificity. Full length PEPP2 associated with both plasma membrane and microtubules. The membrane-associated PEPP2 nucleated at cell-cell contacts and the leading edge of migrating cells. Overexpression of PEPP2 increased membrane microviscosity,...

Allelochemicals of Alexandrium minutum: Kinetics of membrane disruption and photosynthesis inhibition in a co-occurring diatom ArchiMer
Long, Marc; Peltekis, Alexandra; González-fernández, Carmen; Hégaret, Helene; Bailleul, Benjamin.
Allelopathy is an efficient strategy by which some microalgae can outcompete other species. Allelochemicals from the toxic dinoflagellate Alexandrium minutum have deleterious effects on diatoms, inhibiting metabolism and photosynthesis and therefore give a competitive advantage to the dinoflagellate. The precise mechanisms of allelochemical interactions and the molecular target of allelochemicals remain however unknown. To understand the mechanisms, the short-term effects of A. minutum allelochemicals on the physiology of the diatom Chaetoceros muelleri were investigated. The effects of a culture filtrate were measured on the diatom cytoplasmic membrane integrity (polarity and permeability) using flow-cytometry and on the photosynthetic performance using...

Ameliorative Effects of Thymoquinone on Sperm Parameters and Testosterone Level of Nicotine-Treated Sprague Dawley Rats BABT
Rosli,Farah Dayana; Hashim,Noor Hashida; Osman,Khairul; Ibrahim,Siti Fatimah; Kabir,Nurul.
Abstract Thymoquinone (TQ), the main constituent of the volatile oil derived from Nigella sativa has shown pharmacological benefits against various diseases while nicotine is an active component in cigarette that is known to be detrimental. This study was conducted to assess the ameliorating effects of TQ on sperm count, membrane, mitochondria and testosterone of nicotine-treated rats. Rats were randomized into four groups: control, nicotine, TQ, and nicotine with TQ. Nicotine (5 mg/kg bwt/day) was subcutaneously injected for 30 days to induce damaging effects on sperm and testosterone level. Rats were force-fed with TQ (5 mg/kg bwt/day) for the following 30 days. Sperm count was reduced in the nicotine group (26.72 ± 1.64 106/mL) but showed a...

Chill-induced changes in fatty acid composition of tonoplast vesicles from hypocotyls of Vigna unguiculata (L.) Walp. Braz. J. Plant Physiol.
Oliveira,Luciana M. N. de; Sobreira,Alana C. de M.; Monteiro,Fernando de P.; Melo,Dirce F. de.
The present study was undertaken to investigate the changes induced by chilling on fatty acid composition of tonoplast vesicles from hypocotyls of Vigna unguiculata (L.) Walp. The 7 day-old control seedlings were grown at 25°C while treated seedlings were submitted to low temperatures (10°C and 4°C) for 4 days after 3 days germination. The chilling stress resulted in a differential inhibition of plant growth at 10°C and 4°C. Following chilling at 10°C and 4°C, the rate of unsaturated to saturated fatty acids increased under chilling stress. Our results suggest that the increase of unsaturated fatty acids content as well as the higher rate unsaturated/saturate fatty acidsmight be used by V. unguiculata plants as an adaptation mechanism likely to maintain...

Effect of a mono-specific algal diet on immune functions in two bivalve species - Crassostrea gigas and Ruditapes philippinarum ArchiMer
Delaporte, Maryse; Soudant, Philippe; Moal, Jeanne; Lambert, Christophe; Quere, Claudie; Miner, Philippe; Choquet, Gwenaelle; Paillard, Christine; Samain, Jean-francois.
The impact of diets upon the fatty acid composition of haemocyte polar lipids and consequently upon immune parameters has been tested in the oyster Crassostrea gigas and the clam Ruditapes philippinarum. Oysters and clams were fed each of three cultured algae: Chaetoceros calcitrans, which is rich in 20:5(n-3) and 20:4(n-6) and poor in 22:6(n-3) fatty acids; T-Iso (Isochrysis sp.), which is rich in 22:6(n-3) and deficient in 20:5(n-3) and 20:4(n-6); and Tetraselmis suecica, which is deficient in 22:6(n-3) and contains only small amounts of 20:5(n-3) and 20:4(n-6). Fatty acid composition of haemocyte polar lipids was greatly affected by the diet. Oysters and clams fed C. calcitrans maintained a higher proportion of 20:5(n-3) and 20:4(n-6) in their haemocyte...

Leukocyte adhesion - a fundamental process in leukocyte physiology BJMBR
Gahmberg,C.G.; Valmu,L.; Tian,L.; Kotovuori,P.; Fagerholm,S.; Kotovuori,A.; Kantor,C.; Hilden,T..
Leukocyte adhesion is of pivotal functional importance. The adhesion involves several different adhesion molecules, the most important of which are the leukocyte ß2-integrins (CD11/CD18), the intercellular adhesion molecules, and the selectins. We and others have extensively studied the specificity and binding sites in the integrins and the intercellular adhesion molecules for their receptors and ligands. The integrins have to become activated to exert their functions but the possible mechanisms of activation remain poorly understood. Importantly, a few novel intercellular adhesion molecules have been recently described, which seem to function only in specific tissues. Furthermore, it is becoming increasingly apparent that changes in integrins and...

Pre-purification by membrane filtration of paralytic shellfish toxins from Alexandrium minutum dinoflagellate ArchiMer
Balti, Rafik; Brodu, Nicolas; Zhang, Jiaxuan; Amzil, Zouher; Drouin, Delphine; Sechet, Veronique; Masse, Anthony.
The Paralytic shellfish neurotoxins (PST) are of increasing interest for biomedical applications. The chemical synthesis is often complex and expensive that’s why the purification by membrane filtration of PST from Alexandrium minutum dinoflagellate was investigated. Disrupted micro-alga cells by ultrasonic treatment were diafiltred to let pass toxins through an ultrafiltration membrane. Then, the mean permeate was concentrated and diafiltrated by nanofiltration. Mean permeate fluxes equal to 187, 135 and 135 L.h–1.m-2 were obtained during the first diafiltration, the concentration step and the final diafiltration respectively. Up to 57 % (mol/mol) and 78 % (mol/mol) of organic matters and salts were removed respectively. Divalent ions were sparsely...

Sanitary aspects and technological challenges of whole milk microfiltration at low temperatures PAB
Fagnani,Rafael; Mexia,Marisa Marroni; Puppio,Ana Amélia Nunes; Battaglini,Ana Paula Pavão.
Abstract: The objective of this work was to evaluate the effects of whole milk microfiltration at low temperatures on bacterial counts and on its shelf life. The microfiltration process was evaluated at two temperatures (30 and 50ºC) and compared with the slow pasteurization process. Both slow pasteurization and microfiltration reduced the initial counts of aerobic mesophilic and psychrotrophic bacteria, as well as total coliforms in whole milk. Microfiltration at 50ºC was as effective as the pasteurization process, since it reduced the initial count of aerobic mesophilic bacteria in 4.4 log cycles; increased the product's shelf life, which reached 30 days without exceeding 1,000 CFU mL-1; and eliminated coliform counts at the temperatures of 35 and 45ºC,...

Sustainable alternative for the food industry: converting whey and orange juice into a micro-filtered beverage Scientia Agricola
Fagnani,Rafael; Puppio,Ana Amélia Nunes; Zanon,Emely Osti.
ABSTRACT: Enhancing industrial sustainability by converting whey into alternative high value-added products is a scientific trend in food science and technology. However, without other ingredients, rennet, or sour, whey has an unappetizing flavor. This sensory challenge can be overcome by blending it with citrus flavor from orange juice. This study assessed a micro-filtered beverage from whey and orange juice without enzymatic treatment. Four formulations (27:10, 8.6:10, 2.4:10 and 1:10 v/v whey:juice ratio) were processed through a 1.4 μm microfiltration system with four different transmembrane pressures (1, 2, 3, and 4.15 bar) and then stored at 5 °C for 28 days. The micro-filtered beverage was analyzed for physicochemical, sensorial and microbial...
